This screening study investigated the potential of two inorganic Mn-containing Materials (i.e.
MnCl2 and Manganese metal Powder to affect the steroidogenic pathway, using the human H295R cell line in accordance with OECD Guideline 456.
A single steroidogenesis assay experiment was performed whereby the test materials were
tested at seven concentrations together with the positive control inducer forskolin and positive
control inhibitor prochloraz.
MilliQ-water (MQ) was used as vehicle for the test materials and dimethyl sulfoxide (DMSO)
was used as vehicle for the control materials.
H295R cells were exposed for 48±2 hours to the vehicle, test materials and positive controls.
A single valid steroidogenesis screening experiment was performed. Fold-changes ≥ 1.5 in
estradiol and testosterone concentrations compared to the solvent control (i.e. a 1.5-fold
increase or 1/1.5 = 0.67-fold decrease) did not occur at any of the evaluable concentrations
for both test materials. Overall, the results for MnCl2 and Mn metal powder met the criteria of a
negative response in the H295R steroidogenesis assay. The NOEC (No Observed Effect
Concentration) was 100 μM (based on this 1.5-fold threshold), which was the highest noncytotoxic concentration tested.
